The three drugs dexamethasone, promethazine and nordihydroguaiaretic acid were observed to sensitize mice to an otherwise-tolerated dose of cycloheximide. It was suggested that the three agents induced fatal susceptibility to cycloheximide through the action of the gut microbiota. It was also proposed that the three drugs impair the hepatic mechanism which removes endogenous lipopolysaccharide from the portal vein. The resulting systemic distribution of lipopolysaccharide was described before to render mice fatally susceptible to subsequent cycloheximide challenge.
