The rat caecal microbiota is able to deacylate phenacetin and certain related compounds to the corresponding primary amines in vitro, while other N-acylated drugs resisted microbial deacylation. When rat caecal contents were incubated with phenacetin under anaerobic conditions, the deacetylation reaction involved in forming of p-phenetidin could be demonstrated.
